Molecular root of 'exhausted' T cells in chronic viral infection

When you get an acute infection, such as influenza, the body generally responds with a coordinated response of immune-cell proliferation and attack that rapidly clears the pathogen. Then, their mission done, the immune system stands down, leaving a population of sentinel memory cells to rapidly redeploy the immune system in the event of reinfection.

This is why vaccination works, and it's why, in theory at least, people who have had the chicken pox once will never get it again.

But what about chronic infection? In the case of such pathogens as hepatitis C, HIV, and malaria, the body and the pathogen essentially fight to a prolonged stalemate, neither able to gain an advantage. Over time, however, the cells become "exhausted" and the immune system can collapse, giving the pathogen the edge.

Now, a new study by researchers at the Perelman School of Medicine, University of Pennsylvania, is showing just how that happens. The findings also suggest a novel therapeutical approach that might be used to shift the balance of power in chronic infections. The study appears in the November 30 issue of Science.

The team, led by E. John Wherry, PhD, associate professor of Microbiology and Director of the Institute for Immunology, used a mouse model of chronic viral infection to map the T-cell response that arises when the immune system is on an extended war footing. They found that two distinct classes of virus-specific CD8+ T cells ? one expressing high levels of the protein T-bet, the other expressing high levels of the protein Eomes, work together to keep the infection in check.

Specifically, they found that the two cell populations appear to have a progenitor-mature cell relationship. The T-bet-expressing cells appear to function as the progenitor cells ? that is, stem cells. These cells divide both to regenerate and maintain the pool of virus-specific T cells. But they also divide and differentiate to form mature, terminally differentiated Eomes-expressing cells. These cells are more effective at fighting the virus itself, but cannot replicate.

"There's a balance, an equilibrium, which allows you to maintain control over the infection but is insufficient to give you complete clearance," Wherry explains.

These two cell subpopulations tend to confine themselves to different anatomic regions in the infected animals, the researchers found. T-bet-positive cells were found in the blood and spleen, whereas Eomes cells were found in the liver, bone marrow, and gut.

Loss of either subpopulation, which the researchers modeled by deleting one or the other protein, reduces the immune system's ability to fight the infection, leading to a shift in favor of the pathogen.

According to Wherry, these data can help explain the gradual loss of virus-specific T cells observed in such chronic infections as hepatitis C.

"Our data suggest the reason for loss of immune control during some chronic infections is that the long-term pressure on this progenitor-mature cell relationship depletes the progenitor pool," he says.

What's more, the study suggests new therapeutic avenues that can be used to fight, or at least better control, chronic infections. For instance, he says, "If we can maintain these progenitor cells longer, or coax the terminal progeny to divide further, we may be able to shift the balance and maintain control of the infection," he says.

Wherry's lab is now studying candidate molecular pathways to determine their efficacy in controlling, and perhaps modulating, these two T-cell populations.

understanding facebook edgerank Yes, You Have to Pay to Reach Some of Your Facebook FansI promised myself that I wouldn?t write another EdgeRank or Reach controversy article for a while, but I couldn?t resist?

Now that Facebook has tweaked EdgeRank, the common refrain is that brands are being forced ? blackmailed even ? to pay to reach their Fans.

My response: Well, yeah. Sorta.

And my follow-up: That makes Facebook a better place.

This deserves an explanation. First, let?s understand how Facebook EdgeRank works?

The People You Reach


Today, when you write your historically, insanely awesome Facebook post, you?re going to reach some people. Let?s try to break down how that is going to happen.

This historically, insanely awesome piece of content is going to reach?

  • Fans who regularly engage with you and who were online when you posted,
  • Fans who sometimes engage with you and were online when you posted, and
  • People who saw the engagement of friends with your awesome content.

The People You Don?t Reach

The following people will be extremely unlikely to see your content:

  • People who have hidden your content or marked it as spam,
  • Fans who have never clicked on or engaged with your content,
  • Fans who have rarely clicked on or engaged with your content, and
  • Fans who weren?t online within a couple of hours of your post.

The Other Reach Variables


This is if your content is historically, insanely awesome. If it?s average ? meaning that it doesn?t get a ton of clicks, comments, likes or shares ? expect to reach fewer people.

Who you reach then will be dependent upon a combination of?

  • How often they engage with you,
  • How often they engage with that type of content, and
  • How often others are engaging with that content.

Reaching the Rest


Is that unfair? Not really. It?s Facebook surfacing your content to the people most likely to want to see it.

Think that?s BS? Not according to the results I?ve seen ? both on my Page and globally. Reach does seem to be down a bit, but engagement remains steady. That can only be the case if Facebook has made the News Feed more relevant and efficient.

Let?s use an elementary example. Before, I reached 100 people to get 20 forms of engagement. Now I?m reaching 90 people to get the same 20 forms of engagement.

Should I feel wronged for not reaching the 10 invisible people?

There?s something that needs to be said here that brands do not want to admit: Some of your Fans simply do not care if they hear from you. Even stronger, some don?t want to hear from you at all.

Don?t assume that the ?Like? is an implied subscription to see all of your content. People ?Like? for different reasons. Sometimes they want to see your stuff. Sometimes it?s just as an expression.

But don?t think it?s not perfectly clear how they feel about you. If they wanted to see your content, they would be engaging with it. And when Facebook sees they don?t care about your content, Facebook stops showing it to them.

Maybe they don?t want to hear from you in general. Maybe the tepid response to your post is a cue to Facebook that most won?t want to hear from you. But it?s something you need to understand and accept.

The ?Blackmail?


So, you need to pay a few bucks to reach those people? Gotta shell out some green to invade the News Feeds of people even though your content was crap? Forced to pony up to get in front of someone who hasn?t once engaged with your content?

It actually makes sense to me. And I think it makes Facebook a better place.

If you aren?t reaching the number of people you think you should reach ? if the engagement is also low ? you have two choices: 1) Learn and adjust, or 2) Pay to reach those less willing to be reached.

You could yell and scream that Facebook is screwing you. But I don?t think that will get you very far.

Personally, I wouldn?t pay to promote a post that isn?t getting engagement. That?s your sign that it?s a waste of money.

I often have people tell me that a Facebook ad ?failed? because they spent $100 and didn?t get much of anything in terms of engagement or conversions. But they?re missing the point.

You paid to reach more people. Those people may not have wanted to be reached. You can?t pay to force them to engage. If they don?t engage, it?s a sign that your content was a failure. Not your ad.

So, is it blackmail to force you to reach people who weren?t online, don?t engage with your content or likely wouldn?t be interested based on initial response?

No way. It?s understanding what users want. It?s controlling the flow of stuff they don?t want. And it?s profiting if you want to override that control.

I'm not so sure this is the best thing in the world, 100%, as you don't have an option to turn it off. I like the idea of inviting people to share their thoughts, and mixing in real names and pictures if chosen by the user. That way, people know what reviews they might want to trust over other ones that are anonymous and thus, perhaps shady. The company seems to be choosing quality over quantity, though.

Q: We lost our Soft-Coated Wheaton Terrier, and it's still very difficult to talk about and deal with the loss. Poor Gracie, our other dog, has also had a tough time, and it's been heartbreaking. I think Gracie is still waiting for McGee to come home. There's no way to tell her that's not going to happen. How can we help Gracie through this difficult period? -- S.O., Chicago, IL.

A: I'm very sorry for your loss. I -- and many other readers -- have felt a similar hole in our hearts.?We don't really know what surviving pets are thinking when it appears for all the world as if they're grieving. They may be, or they may be merely picking up on our sorrow. It could be they're only responding to a sudden change in the household - or the changes may be a combination of all those things. Personally, I'm convinced that pets (who we know, for a fact, feel emotion) can grieve. But why do some pets appear absolutely unaffected by the loss of a best pal? No one knows.

"On average, people actually take about two years to grieve the loss of a loved one," says Sue Yellen, Glenview, IL-based clinical psychologist and chicagonow.com blogger. "I suspect many pets do grieve, though we don't understand that grieving period."

She adds, "It's a difficult balance, you want to give attention to people who are grieving, and I believe the same is true for grieving pets. However, you also don't want to reward sad behavior too much."

Try to keep a regular schedule for Gracie's activities, so there's a consistent structure to her life. Feed her and take her for walks at about the same time you always have. Playing with a dog's favorite toy is probably the best antidote (for dogs who enjoy play), and the exercise is a great outlet for both you and your pet.

Wall St edges down after recent rally; retailers weigh

NEW YORK (Reuters) - Wall Street slipped on Monday, pulling back from last week's gains, as retailers fell on concerns about heavy discounts at the start of the U.S. holiday shopping season and the overhang of the "fiscal cliff" kept investors wary of making big bets.

The Nasdaq outperformed to close higher, led by gains in eBay and as Apple continued its bounce back.

The Standard & Poor's 500 cut most of its losses during the session and managed to stay above the psychologically important 1,400 level. It also remained above the 200-day moving average, maintaining its long-term uptrend.

The S&P 500 consumer discretionary index <.gspd> fell 0.5 percent after the start of the holiday shopping season over the four-day Thanksgiving weekend. Target , one of the largest retailers by market value, fell 2.6 percent.

"The concern is big retailers are discounting so much, sales look better, but at what cost?" said Angel Mata, managing director of listed equity trading at Stifel Nicolaus Capital Markets in Baltimore.

Bucking the retail trend, shares of eBay closed at their highest in almost eight years, rising 4.9 percent to $51.40, as the online marketplace notched strong sales on "Cyber Monday." Amazon gained 1.6 percent to $243.62.

The White House threw cold water on a proposal of avoiding the looming "fiscal cliff" of spending cuts and tax highs by limiting tax deductions and loopholes, instead of allowing tax rates to rise for the richest Americans.

Investors are hoping for advances in talks over the $600 billion in spending cuts and tax hikes scheduled to begin next year, which threaten to drag the U.S. economy back into recession.

Indications of progress in talks, or just political willingness to negotiate, contributed to the market's recent rally. Major indexes last week gained 3 to 4 percent, with the Dow above 13,000 and the S&P above 1,400 for the first time since November 6.

Those gains represented a turnaround from recent losses founded on worries about Washington's ability to solve budgetary problems.

The Dow Jones industrial average <.dji> fell 42.31 points, or 0.33 percent, to 12,967.37. The S&P 500 <.spx> dropped 2.86 points, or 0.20 percent, to 1,406.29. The Nasdaq Composite <.ixic> gained 9.93 points, or 0.33 percent, to 2,976.78.

About 5.2 billion shares changed hands on the New York Stock Exchange, the Nasdaq and NYSE MKT, below the daily average so far this year of about 6.49 billion shares.

On the NYSE, roughly 13 issues fell for every 10 that rose, and on Nasdaq nearly six rose for every five that fell.

In the other major worry for the market, euro zone finance ministers and the International Monetary Fund made their third attempt in as many weeks to agree on releasing emergency aid for Greece, with policymakers saying a write-down of Greek debt is off the table for now.

"There's no catalyst to continue the rally we saw last week, though Greece would have been important if we weren't dealing with the fiscal cliff," Stifel Nicolaus' Mata said.

Shares of Knight Capital Group Inc jumped 13.3 percent to $2.82 following reports that rivals might be preparing to bid for part or all of the trading firm.

Apple Inc has asked a federal court to add six more products to its patent infringement lawsuit against Samsung Electronics , including the Samsung Galaxy Note II, in the latest move in an ongoing legal war between the two companies. Apple shares were up 3.2 percent at $589.53.

GOP opposition to diplomat Rice begins to crack

WASHINGTON (AP) ? Republican opposition to the possible nomination of Susan Rice to be secretary of state is showing signs of cracking.

Arizona Sen. John McCain now says he'd be willing to meet with Rice, the current U.N. ambassador, and consider her much-scrutinized explanation about how the Sept. 11 attack on the U.S. Consulate in Benghazi, Libya, unfolded.

McCain has said GOP senators would do "whatever is necessary" to block Rice if President Barack Obama were to nominate her to replace Hillary Rodham Clinton at the State Department.

On Sunday, McCain was asked on "Fox News Sunday" if Rice could change his mind. McCain said "sure" and that he'd like to meet with her.

McCain and other Republicans have accused the White House of distorting the deadly Benghazi attack for political gain.

Your Brand Matters

Any business needs to consider its branding regardless of its niche, offering or brand recognition (if you happen to enjoy a monopoly on supplying oxygen to the planet, you can skip this? otherwise, pay attention).

It doesn?t matter if you?re business is Don?s Trading Post in tiny Buford, WY, or Google? if you expect to keep the customers you have or gain any new ones, you need to nurture your brand?s reputation. Virtually every business has some brand identity ? even if it?s only established in the mind of one customer. It?s important to remember, though, that a brand identity can be either negative or positive.

Coca Cola probably enjoys a positive brand image with lovers of their iconic cola. But it?s likely less positive with Pepsi lovers. That?s no fault of the company, of course. Every consumer has his preferences, and as much as you try, you can never please everyone in all regards.

However, if you consider customer service to be a major portion of your branding effort, you can come mighty close.

On the flip side of that coin, there are instances of businesses going far beyond reasonable expectations to ensure their customers are aware of how important they are. Such stories can be inspiring to consumers and employees alike and just like with negative occurrences, one such incident can yield results that last years.

Setting the Company Culture

Whether a business is a one-man operation or has thousands of employees, an attitude of providing exemplary customer service starts at the top. The CEO and Board of Directors need to stress the importance of taking care of customers, and every level of management, supervision and production, right down to the night watchman (that poor lonely fellow that never gets to see anyone), needs to live by it.

Companies that go the extra mile to take care of their customers reap benefits, sometimes in big ways. Employees that display such willingness to go the extra mile should be rewarded, too. It doesn?t have to be a huge cash award or a trip to the Bahamas. Sometimes recognition alone can be enough to make it clear to others that customer service isn?t just a buzz-word to the company?s management ? it?s a mission.

Recognizing and rewarding the sort of behavior you want to see from your employees is just common sense. It encourages more of the same from others, and it definitely discourages less favorable attitudes from any that haven?t ?seen the light?. Nobody wants to be the one seen as guilty of dropping the ball on an important company value.

Building your brandBrand is EVERYTHING, especially when you don?t have it yet ? and even more so when what you have is negative. The perception that consumers (and potential consumers) have of your company?s brand is centered around your reputation. Whether it deals with quality, price, service or delivery, it all boils down to value. And of those four, service is the one that involves the least cost and is the most easily implemented and maintained.

It?s also the one that can trump all the others. If you?re not focusing on building your brand, you?re missing the boat. And if you?re trying it do it without focusing on service, you are missing a great business opportunity ? one you may not get another shot at.

Rolling Stones pounce on early hits at London show

LONDON (Reuters) - The Rolling Stones hit the stage at London's O2 arena on Sunday with a roar of approval from a sea of silver-haired fans as they tore into old favorite "I Wanna Be Your Man".

A swaggering Mick Jagger dressed in sparkling silver and black rolled back the years with the 1960s hit as he bounced onto a stage shaped in the red lips logo that is the calling card for one of rock and roll's most enduring bands.

Lead singer Jagger, guitarists Keith Richards and Ronnie Wood and drummer Charlie Watts are all in their 60s and early 70s, but have promised a two-hour show that will stun the crowd and include former band members Bill Wyman and Mick Taylor as well as R&B singer Mary J. Blige and guitar great Jeff Beck.

"Everybody all right there in the cheap seats?" Jagger asked in reference to a controversy over the price of tickets. "They're not really cheap though are they? That's the trouble."

A sellout crowd of some 20,000 people was expected in spite of widespread complaints from fans at ticket prices that ranged from 95 pounds ($150) to up to 950 pounds for a VIP seat at the first of five concerts celebrating the band's 50th anniversary.

Costs went far higher on secondary ticketing websites, although by Friday eBay was offering several seats to Sunday's show at below face value and there were places still officially available at around 400 pounds apiece.

There has been talk of a wider tour, but for the time-being the only confirmed concerts are the five that have been announced. Two will be played at the O2 Arena in London, the first was under way on Sunday and another on Thursday with three others in the United States next month.

The flamboyant veterans behind a string of hits including "(I Can't Get No) Satisfaction", "You Can't Always Get What You Want" and "Jumpin' Jack Flash" have defended the prices, saying the shows are expensive to put on, although specialist music publication Billboard reported the band would earn $25 million for the four shows first announced. A fifth was added later.

BURST OF ACTIVITY

The concerts are the culmination of a busy few months of events, rehearsals and recordings to mark 50 years since the blues-infused rockers first took to the stage at the Marquee Club on London's Oxford Street in July, 1962.

There has been a photo album, two new songs, a music video, a documentary film, a blitz of media appearances and a handful of warm-up gigs in Paris.

The O2 Arena was where another top band of the 1960s and 70s, Led Zeppelin, staged an eagerly awaited one-off reunion in 2007, and while the Stones have appeared together far more regularly, it is their first arena performance in six years.

One factor behind the long break has been Wood's struggle with alcohol addition, according to Rolling Stone magazine, while Jagger and Richards also fell out over comments the guitarist made about the singer in a 2010 autobiography.

"We can't get divorced - we're doing it for the kids!" joked Richards in a recent interview after apologising to Jagger.

While the rock and roll excesses of the swinging 60s and 70s are in the past for the band, and their very best songs may be behind them, music critics praised their recent single "Doom and Gloom" from the "GRRR!" greatest hits album just released.

And there have been hints from the band that the five gigs which wind up at the Newark Prudential Center on December 15 may not be the end of their reunion.

"Once the juggernaut starts rolling, it ain't gonna stop," Richards told Rolling Stone. "So without sort of saying definitely yes - yeah. We ain't doing all this for four gigs!"
